            尿激酶型纖溶酶原激活物(Urokinase type Plasminogen Activator (uPA) )，uPA是一種絲氨酸蛋白水解酶。小鼠uPA基因位于第10條染色體長臂上，其長度為6.5KD，由11個外顯子構(gòu)成，可轉(zhuǎn)錄成24KB的成熟的mRNA，初始翻譯產(chǎn)物為前尿激酶原（含431個氨基酸殘基），除去20個氨基酸殘基的信號肽后即為細(xì)胞分泌的尿激酶原（pro-uPA），在158LYs位處劈裂，則成為由二硫鍵連結(jié)的雙鏈活性型結(jié)構(gòu)，即20×103的A鏈和34×103的B鏈。C端的A鏈包括一個Kringle區(qū)和表皮生長因子（EGF）功能區(qū)，表皮生長因子功能區(qū)能介導(dǎo)與特異的受體結(jié)合，去掉此功能區(qū)，其仍具有激活纖溶酶原作用，,但卻無與受體結(jié)合的能力。C端的B鏈有絲氨酸蛋白酶功能區(qū)，為uPA的活性中心。uPA的合成與分泌受諸多因素影響，如原癌基因V-Src、Ras、細(xì)胞因子TNF、INF、IL-1、TGF-β、IGF-1、FGF、基質(zhì)金屬蛋白酶等。一般說來，uPA主要在生理、病理?xiàng)l件下與細(xì)胞分化、遷移、組織重建、細(xì)胞周圍基質(zhì)降解有關(guān)。
