            Nanoprobes offers a new line of Alexa Fluor® 647 FluoroNanogold™ antibody and streptavidin conjugates labeled with both Alexa Fluor® 647 dye and the 1.4 nm Nanogold® particle. The dual labels are covalently attached to the antibody or streptavidin, enabling both fluorescence and electron microscope observation of the same structure in a single labeling procedure. The Alexa Fluor® 647 dye is a bright, far-red–fluorescent dye with excitation ideally suited for the 594 nm or 633 nm laser lines, and is useful in stochastic optical reconstruction microscopy (STORM). It can also be used in two-photon excitation (TPE) microscopy . The presence of the 1.4 nm Nanogold® particle allows electron or light microscope detection with and without silver or gold enhancement . The presence of both the fluorescent and the gold labels make correlative microscopy study possible.

             

            PRODUCT INFORMATION

             

            Alexa Fluor®647 FLUORONANOGOLD™ is a unique, dual-purpose probe1 . Alexa Fluor® 647 FLUORONANOGOLD™ conjugates consist of affinity-purified IgG molecules, Fab' fragments or streptavidin conjugated to both Alexa Fluor® 647 dye and the 1.4 nm NANOGOLD® particle.2 The structures of Alexa Fluor® 647 FLUORONANOGOLD™ IgG and Fab' conjugates are shown in Figure 1. In the fluorescence microscope, these probes may be used just like conventional fluorescently-labeled antibodies,3 while in the electron microscope they are visualized in exactly the same manner as for NANOGOLD® conjugates4 . The combination of fluorescence and electron microscopy, which is referred to as correlative microscopy13, allows imaging of the same structure in both microscopes. The covalent label linkage is stable indefiniy, and the attachment of NANOGOLD® at a hinge thiol site ensures maximum preservation of native immunoreactivity. These reagents are supplied at a concentration of 0.08 mg/mL of antibody dissolved in 20 mM phosphate buffered saline (150 mM NaCl) at pH 7.4 (PBS), with 0.1 % BSA and 0.05 % sodium azide as preservatives. Alexa Fluor®647 FLUORONANOGOLD™ conjugates should be stored at 2-8°C. DO NOT FREEZE. PROTECT FROM LIGHT.

            Congratulations on your acquisition of a unique dual cytochemical labeling reagent: Alexa Fluor® FluoroNanogold™-Streptavidin. This unique streptavidin probe contains both the 1.4 nm Nanogold® particle and Alexa Fluor®, both covalently bound, and enables both fluorescence and electron microscopic observation of a sample stained using a single labeling procedure. This probe is smaller than a whole IgG molecule, does not aggregate, and fluorescence quenching due to the gold particle is low. Alexa Fluor® has been found to have significant advantages over fluorescein or rhodamine: it is brighter and quenches less readily.

These reagents are supplied at a concentration of 0.08 mg/mL of streptavidin dissolved in 20 mM phosphate buffered saline (150 mM NaCl) at ph 7.4 (PBS), with 0.1% BSA and 0.05% sodium azide as preservatives. The degree of labeling for the conjugate is typically one Nanogold® particle and 2-3 fluorophores per streptavidin molecule, and the exact degree of labeling is indicated on the product specification sheet. FluoroNanogold™ conjugates should be stored at 2-8°C. DO NOT FREEZE.
