| I. Background | |
| 白細(xì)胞介素-12 （IL-12） 是一種異二聚體促炎細(xì)胞因子，可調(diào)節(jié) T 細(xì)胞和自然殺傷細(xì) 胞反應(yīng)，誘導(dǎo)干擾素-γ （IFN-γ） 的產(chǎn)生，促進(jìn)輔助性 T 細(xì)胞 1 （TH1） 細(xì)胞，是先天抵 抗和適應(yīng)性免疫之間的重要紐帶。 IL12 家族還包括 IL23、IL27、IL35 和 IL39 。IL-12 和 IL-23 由人和小鼠的抗原呈遞細(xì)胞（樹(shù)突狀細(xì)胞和組織內(nèi)巨噬細(xì)胞）產(chǎn)生，以響應(yīng)與宿主防 御和傷口愈合相關(guān)的外源或內(nèi)源性信號(hào)，對(duì) CD8 T 和 NK 細(xì)胞的募集和效應(yīng)功能至關(guān)重要。IL-12 可以中和 CD8 T 細(xì)胞上的負(fù)調(diào)節(jié)受體的信號(hào)傳導(dǎo)。例如，IL-12 下調(diào) CD8 T 細(xì)胞上的 PD-1 和 IFNγ表達(dá)，保護(hù)腫瘤浸潤(rùn)的 CD8 T 細(xì)胞免受 IFN γ誘導(dǎo)的細(xì)胞死亡。抗 PD1 激 活抗腫瘤免疫需要 IL-12 介導(dǎo)的 T 細(xì)胞和樹(shù)突狀細(xì)胞之間的串?dāng)_，從而能夠殺死 CD8 T 細(xì) 胞介導(dǎo)的腫瘤細(xì)胞。 IL-12 由 IL-12p35 亞基相連 IL-12p40 亞基組成，由 IL-12 受體（IL-12R）組成的異二 聚體發(fā)出信號(hào)，后者包含 IL-12Rβ1 和 IL-12Rβ2 亞基。IL-12 刺激 JAK2 和 TYK2 活性，導(dǎo) 致信號(hào)轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄激活因子（STAT）家族成員 STAT1、STAT3、STAT4、STAT5 的磷酸化。 | |
