DR3 Effector Reporter Cell
| I. Background | |
| TL1A（腫瘤壞死因子樣細(xì)胞因子 1A）是 TNF 蛋白超家族（TNFSF）的成員，是一種 II 型跨膜蛋白，與 TNF 家族的其他成員類似，TL1A 形成穩(wěn)定的三聚體。它以膜結(jié)合形式 （mTL1A）存在，也可能被基質(zhì)金屬蛋白酶裂解并釋放為可溶的、全功能的蛋白質(zhì) （sTL1A）。白質(zhì) （sTL1A）。 TL1A 的功能性受體是 DR3（死亡結(jié)構(gòu)域受體 3），是一種 I 型跨膜蛋白，它含有四個(gè) 半殘基、兩個(gè)潛在的 N 連接糖基化位點(diǎn)的胞外域、一個(gè)跨膜結(jié)構(gòu)域和一個(gè)帶有死亡 結(jié)構(gòu)域的胞內(nèi)域。sTL1A/DR3 相互作用可觸發(fā)兩種不同的信號(hào)通路，分別引起炎癥和細(xì)胞 凋亡 。 DR3 的死亡結(jié)構(gòu)域先與胞漿中的銜接蛋白 TNFR 相關(guān)死亡結(jié)構(gòu)域蛋白 (TRADD) 結(jié) 合，進(jìn)而募集 TNFR 相關(guān)因子 2 (TRAF2) 和受體相互作用蛋白 1 (RIP1)，這些復(fù)合物激活 MAPKs (ERK、p38 和 JNK)、NF-κB 以及效應(yīng)激酶 PI3K 信號(hào)轉(zhuǎn)導(dǎo)，最終調(diào)控促炎基因表 達(dá)，參與免疫相關(guān)疾病的發(fā)生。 | |
