Stem121：抗人類細胞質(zhì)蛋白的小鼠抗體
1. Stem121是什么？
· Stem121是小鼠來源的單克隆抗體，它特異性地識別人類細胞（特別是人類神經(jīng)細胞）細胞質(zhì)中的某個抗原。
· Stem121在反應(yīng)屬性上有兩個主要特點：
1) 它所識別的抗原不在細胞核內(nèi)，可以特異性的識別細胞質(zhì)內(nèi)的抗原；
2）它與小鼠、大鼠、猴子細胞的細胞質(zhì)沒有反應(yīng)性。
2. Stem121的應(yīng)用
· 小鼠、大鼠、猴子是目前干細胞治療研究者常用的模式動物。通常，研究者將人類的神經(jīng)干細胞注射進入這些模式動物的腦部，然后觀察這些細胞是否可以在動物的腦部存活、生長、分化、遷移。因此，必須可靠而且準確地示蹤在動物體內(nèi)的人類神經(jīng)干細胞。
· Stem121抗體是可以對人類細胞細胞質(zhì)進行標記的抗體。
· 對于輸入的人神經(jīng)干細胞，既標記細胞核又標記細胞質(zhì)，意味著更加可靠、更加準確。如下圖所示，人類的神經(jīng)干細胞被注射進入大鼠的腦部，藍色標記是某種識別人類細胞的細胞核的抗體，綠色標記是識別人類細胞細胞質(zhì)的Stem121抗體，紅色標記是識別人類細胞某種發(fā)炎癥狀的抗體。在A部分的左圖左下角有個箭頭指示的細胞，具備藍色與綠色標記，這說明這個細胞確定是人類細胞而非大鼠細胞；右圖右下角的箭頭指示同一個細胞，沒有綠色標記，說明這個人類細胞并沒有出現(xiàn)發(fā)炎癥狀。而且，如果實驗的設(shè)計目的是觀察人類的神經(jīng)細胞的細胞質(zhì)能否進入到人類或是動物細胞的細胞核里面，那么就需要Stem121對人類細胞質(zhì)進行標記。
