轉(zhuǎn)染試劑那些事兒

適用宿主廣泛，操作簡便，細胞毒性小，轉(zhuǎn)染率高。其中聚乙烯亞胺（PEI）的轉(zhuǎn)染性能比較好。聚乙烯亞胺是一種具有較高的陽離子電荷密度的有機大分子，每相隔兩個碳原子，就是第三個原子都是質(zhì)子化的氨基氮原子，使得聚合物網(wǎng)絡(luò)在任何PH下都能充當有效的“質(zhì)子海綿”體。這種聚陽離子能將各種報告基因轉(zhuǎn)入各種種屬細胞，其效果好于脂質(zhì)聚酰胺，而且細胞毒性低。大量實驗證明，PEI是非常有希望的基因治療載體。目前在設(shè)計更復雜的基因載體時，PEI經(jīng)常做為核心組成成分。
線性聚乙烯亞胺（Line PEI）轉(zhuǎn)染復合物的細胞毒性低，有利于細胞定位，而且轉(zhuǎn)染率*。
Polyscience公司的兩款明星產(chǎn)品線性聚乙烯亞胺（23966-1和24765-1）近年來越來越多的受到研究者們的青睞。其產(chǎn)品被運用到大量的轉(zhuǎn)染實驗中，也出現(xiàn)在大量的文獻中。
Polyscience公司成立于1961年，總部位于賓夕法尼亞州沃靈頓，是一家生產(chǎn)研發(fā)化學產(chǎn)品的公司。該公司的產(chǎn)品種類已經(jīng)累計超過3000種，其產(chǎn)品的特點是體積小，附加值高。

24765-1（更高效的PEI聚乙烯亞胺聚合物轉(zhuǎn)染試劑）
PEI MAX-Transfection Grade Linear Polyethylenimine Hydrochloride (MW40,000)
基本描述：
PolyethylenimineHClMAX,Linear,Mw40,000（PEI40,000）是熱賣產(chǎn)品線性化聚乙烯亞胺PEI25,000的升級產(chǎn)品，不同之處在于1）*水解（去乙?；?/span>2）PEI25，000的鹽酸鹽形式，具更高的水溶特性；3）含更長連續(xù)性乙烯亞胺（ethyleneimine）片段，其質(zhì)子化氮水平比PEI25，000提高11%以上。本品經(jīng)堿中和后，可轉(zhuǎn)化為線性化自由胺（PEI），此時分子量正相當于25,000。
ThomasM，etal（2005）研究數(shù)據(jù)表明，去乙?；?/span>PEI40,000（PolyethylenimineMAX40,000）體外質(zhì)粒DNA轉(zhuǎn)染效率提高21倍，小鼠體內(nèi)靶向效率達到10,000倍，隨之肺部特異療效顯示增強1500倍。

使用方法（僅供參考）
1. Pour ~450 mL of Milli-Q H2O into a 500-mL glass beaker.
2. Add 500 mg of PEI to the beaker with stirring.
3. Add concentrated HCl drop-wise to the solution to pH <2.0.
4. Stir until the PEI is dissolved (~2-3 h). Maintain the pH <2.0 throughout.
Approximately 800 μL of 12 M HCl will be required for full PEI dissolution. There may still be some small fiber particles that will not dissolve.
5. Add concentrated NaOH drop-wise to the solution to pH 7.0.
Approximately 500 μL of 10 M NaOH will be required to neutralize the PEI solution.
6. Pour the solution into a 500-mL glass cylinder. Adjust the final volume to 500 mL with Milli-Q H2O.
7. Filter-sterilize the solution through a 0.22-μm membrane.
8. Store aliquots of the desired volume at -20°C.
