哮喘模型-屋塵螨引起的哮喘
背景簡介：
哮喘是由接觸塵螨、寵物皮屑、花粉或霉菌等過敏原引起的，其特征是氣道受阻和支氣管痙攣。在這些過敏原中，屋塵螨（HDM）是人類最重要的室內(nèi)過敏原。兩種主要的螨蟲種類，屋塵螨（D.pteronyssinus）和粉塵螨（D.farinae）是導(dǎo)致人類過敏源性哮喘的主要變應(yīng)原。
Chondrex提供的屋塵螨和粉塵螨純化提取物，非常適合誘導(dǎo)小鼠過敏性哮喘，用于構(gòu)建與人類過敏性哮喘非常相似的小鼠哮喘動物模型。此外，Chondrex的室內(nèi)塵螨抗體檢測試劑盒、細(xì)胞因子檢測試劑盒和趨化因子檢測試劑盒，也可用于監(jiān)測HDM誘導(dǎo)的哮喘模型的進程，并測定實驗療法的效果。其Der p10檢測試劑盒和胰蛋白酶活性檢測試劑盒，還可用于測定過敏原含量和總蛋白酶活性水平。
1）小鼠過敏性哮喘模型
哮喘新療法的研究需要相應(yīng)的動物模型，盡可能的再現(xiàn)人類哮喘的情形：氣道高反應(yīng)性（AHR）、可逆性氣道阻塞、早期和晚期哮喘反應(yīng)、氣道重塑和嗜酸性粒細(xì)胞增多癥。傳統(tǒng)方法使用卵清蛋白（OVA）誘導(dǎo)小鼠過敏性哮喘。然而，OVA不是臨床上的過敏原，鋁佐劑的體內(nèi)致敏過程與人類由過敏原致敏的過程截然不同。因此，研究人員轉(zhuǎn)向使用臨床相關(guān)的過敏原，如屋塵螨HDM。HDM是一種免疫復(fù)雜的過敏原，可以刺激先天性和適應(yīng)性免疫反應(yīng)。與傳統(tǒng)的OVA誘導(dǎo)哮喘模型相比，HDM誘導(dǎo)的過敏性哮喘更接近于人類過敏性哮喘。HDM誘導(dǎo)的過敏性哮喘模型實驗療法可能會為哮喘患者治療提供新的選擇。
2）屋塵螨（House Dust Mite, HDM,）
HDM提取物，主要來自于屋塵螨和粉塵螨，是人類最重要的室內(nèi)過敏原。它包含20種順序和功能同源的過敏原。其中，Der 1和Der 2是誘導(dǎo)患者免疫反應(yīng)的主要過敏原，也是研究中最常見的過敏原。Der p 1和Der f 1是半胱氨酸蛋白酶，可以切割緊密連接并增加氣道上皮的通透性。這種蛋白水解提升了抗原細(xì)胞識別過敏原的可能性，從而誘導(dǎo)促炎反應(yīng)。哮喘誘導(dǎo)尤其與Der p1蛋白水解活性相關(guān)，可以用胰蛋白酶測定法用顯色底物進行測量。
Der p 2和Der f 2與MD2的氨基酸序列具有高度同源性，MD2是脂多糖（LPS）的共配體，通過TLR4信號激活先天免疫反應(yīng)。除了Der 1和Der 2，HDM還含有其他HDM過敏原，以及細(xì)菌和真菌產(chǎn)物，如LPS和β-葡聚糖。因此，HDM可以在不使用佐劑的情況下激發(fā)適應(yīng)性和先天性免疫反應(yīng)。與需要鋁佐劑的OVA誘導(dǎo)的哮喘模型不同，HDM提取物可單獨用于誘導(dǎo)過敏性哮喘實驗。
實驗流程：
1）屋塵螨誘發(fā)哮喘時間點
目前已發(fā)表多種小鼠誘導(dǎo)HDM哮喘模型的實驗方案。圖1-3顯示了不同劑量HDM下，過敏原激發(fā)過程的示例方案。我們建議根據(jù)您的研究需要建立和優(yōu)化方案，因為HDM的有效劑量取決于小鼠的品系、飼養(yǎng)條件和HDM的質(zhì)量。此外，長期接觸HDM會導(dǎo)致氣道重塑，粘液細(xì)胞密度和AHR增加，即使在停止接觸HDM后，AHR也會保持升高。

2）哮喘模型評估
屋塵螨誘發(fā)哮喘模型可以再現(xiàn)人類過敏性哮喘的情形。但其疾病表型可能因研究條件而異，如小鼠品系、HDM質(zhì)量和挑戰(zhàn)方案。因此，如何選擇疾病評估方法，對于評估疾病嚴(yán)重程度和治療效果尤為重要。
a.小鼠抗屋塵螨抗體分析
建立HDM誘導(dǎo)的哮喘模型需要高濃度的HDM IgE抗體。與傳統(tǒng)OVA模型和人類哮喘相比，HDM模型中的IgE反應(yīng)往往較弱。其中，HDM-IgG1抗體與Th2應(yīng)答相關(guān)，而HDM-IgG2a或IgG2b與Th1應(yīng)答相關(guān)。因此，測量HDM IgE和IgG的抗體濃度有助于判定致敏類型并實施對應(yīng)的治療方案。Chondrex的小鼠塵螨抗體測定試劑盒和小鼠總免疫球蛋白測定試劑盒，可以量化小鼠樣品中HDM抗體的特定亞型和亞類。
b.細(xì)胞因子和趨化因子分析
哮喘誘導(dǎo)會促使Th2相關(guān)細(xì)胞因子（IL-1α、IL-4、IL-5和IL-13）和哮喘相關(guān)IL-33濃度升高。而量化趨化因子濃度，如CXCL16和CCL20、CCL2、CCL5和CXCL8，有助于改善該病的治療方案研究。Chondrex小鼠細(xì)胞因子檢測檢測試劑盒和小鼠趨化因子檢測試劑盒，可滿足您以上檢測需求。
c.BALF中炎性細(xì)胞計數(shù)與特定氣道阻力
在HDM誘導(dǎo)的哮喘模型中，誘導(dǎo)哮喘會增加支氣管肺泡灌洗液（BALF）中中性粒細(xì)胞、嗜酸性粒細(xì)胞、淋巴細(xì)胞和巨噬細(xì)胞的免疫細(xì)胞計數(shù)。此外，氣道高反應(yīng)性（AHR）在疾病進程中也會隨之加重。
